El Draft de 2006, el septuagésimoprimero de la historia, tuvo lugar del 29 al 30 de abril en el Radio City Music Hall de Nueva York. Por vigesimoséptimo año consecutivo, la ESPN retransmitió el evento. Habiendo firmado ya un contrato el día anterior con su nuevo equipo, los Houston Texans, Mario Williams se convirtió en la primera elección del Draft, sorprendiendo así a los comentaristas, quienes predecían que los Texans acabarían eligiendo a Reggie Bush, de USC. Ohio State obtuvo las mayores elecciones en primera ronda (5), mientras que USC obtuvo las mayores elecciones en general (11).

  11. The Chiefs transferred the 117th overall pick to the Jets as compensation for the Jets' permitting Herman Edwards to leave the Jets to become head coach of the Chiefs.
  12. The Miami Dolphins forfeited their fifth-round selection as a result of selecting Manuel Wright in the 2005 NFL Supplemental draft.
  13. This last selection is commonly referred to as Mr. Irrelevant.
